I don't know the acres, but it was one harvest, and it took the harvester about 10.5 hours.

Usually have one when A not growing cotton or having sheep or B While its growing or when i just set up sheep.

How do you find out?

The game will log how many acres you've fertilised/plowed/cultivated/sown/harvested in a session, you can see it in the pause menu at the statistics tab (next to the contracts page). Save and reload the game,don't set any workers running elsewhere and then just do a job on that field. At the end of it the statistics page will show you the session total for that activity, which will be the size of the field.
