After you buy the concentrate, are you required to use the special hose to finish filling, or will any water source work?

Yes, you need to use the special hose included in the pack.
Make sure to have at least 5% fill level of concentrate of whatever type in your sprayer before using the hose. Otherwise it will be overwritten with liquid fertilizer once you use the hose.

Background:
The hose is just a cheap buying station setup to start filling with current fill type in sprayer. I defaults to liquid fertilizer.
Per basegame behaviour, fill levels below 5% will be overwritten by new fill type.
